Title: Deconstruction and restoration of dysfunctional motor circuitries after spinal cord injury
Summary: Spinal cord injury (SCI) results in chronic paralysis and spasticity. Since spasticity is a major impediment to movements and voluntary movements are lost after SCI, we are currently studying a therapeutic strategy to restore movements after SCI based on parallel approaches to prevent maladaptive plasticity of the neural circuitries caudal to the injury and to activate brain centers to appropriately drive those circuits to generate the desired movement. The combinatorial strategy proposed here aims to restore voluntary walking in mouse models of thoracic SCI using behavioral measurements and intersectional mouse genetics combined with pharmacological interference, opto-electrophysiology, and advanced 3D anatomical imaging.
